Comparative diagnostic value of the gel diffusion test and virus isolation in cell culture for detecting equine herpesvirus type 1 (EHV-1)
نویسنده
چکیده
The gel diffusion test was applied direct to field samples from cases of equine abortion, foal mortality and equine paralysis due to equine herpesvirus type 1 (EHV-1). Following polyethylene glycol concentra tion of viral antigen, one line was formed against EHV-1 antiserum after 24 hours incubation at 25°C. Tests carried out simultaneously on 41 field samples showed a high correlation between diagnosis based on this test and virus isolation in primary lamb kidney cell culture. Advantages of gel diffusion over isolation of virus in cell culture were its rapidity and simplicity. The precipitating antigen did not lose activity after heating at 56°C for 30 minutes. A positive reaction was observed with a conta minated sample, which failed to yield virus in the cell culture system.

The herpesviruses infections in equides are caused by five different serotypes of viruses, belonging to family Herpesviridae. The goal of this study was to conduct a seroepidemiological investigation of equine herpesvirus type 1 (EHV-1) and type 4 (EHV-4) in horses and donkeys raised in two provinces and their villages in northeastern Turkey. A total of 666 samples from 423 horses and 243 donke...

An experimental system that permits sensitive and reproducible detection of equine herpesvirus 1 (EHV-1)-specific cytotoxic T-lymphocyte (CTL) activity in the horse was developed. Peripheral blood mononuclear cells (PBMC) collected from immune horses were restimulated in vitro by culture with live EHV-1. Cytotoxic activity against virus-infected, pokeweed mitogen-stimulated lymphoblast targets ...
متن کاملClinical observations and management of a severe equine herpesvirus type 1 outbreak with abortion and encephalomyelitis
Latent equine herpesvirus type 1 (EHV-1) infection is common in horse populations worldwide and estimated to reach a prevalence nearing 90% in some areas. The virus causes acute outbreaks of disease that are characterized by abortion and sporadic cases of myeloencephalopathy (EHM), both severe threats to equine facilities. Different strains vary in their abortigenic and neuropathogenic potentia...

Two antigenically similar subtypes of equine herpesvirus 1 (EHV-1) cause disease in horses. A procedure for rapid differentiation of the two EHV-1 subtypes with monoclonal antibodies was developed. This report describes the first Brazilian equine herpesvirus type 1 (EHV-1) isolation from a single fatal equine herpes myeloencephalopathy case in a mare. The isolation of EHV-1 was confirmed from the first passage of cerebrospinal fluid (CSF) sample in Vero cells by PCR and virus neutralization assay. As virus isolation from CSF is unlikely to be successful, as has been shown in several case ...
